研究方向:分析探针及蛋白质生物技术(化学生物学的分支)

主要基于蛋白和配体相互作用的物理化学原理,用化学计量学方法发掘复杂生物亲和作用系统中多种成分之间的相互作用机制、强度和物质含量相关信息,用于指导设计体外诊断和药物筛选所需生物亲和分析新方法及对应新探针、蛋白配体和蛋白质突变体,并评价其应用潜力;具体涉及如下研究内容:

(1) 体外诊断和药物筛选的生物亲和分析新方法和新探针:以提高分析测试效率和精度为主要目标,基于生物亲和作用分子机制设计所需分析探针和信息发掘过程;探索单通道多组分同步分析所需信息发掘技术和对应的分析探针、单通道多组分同步免疫分析及标记技术、蛋白配体类混合物组合库筛选技术和靶蛋白固定化方法、酶突变体库筛选技术和筛选材料、以蛋白内院色氨酸残基为能量转移供体的均相亲和分析技术、优化探针设计竞争性亲和分析获得线性响应;

(2) 以PDE和GST的同工酶选择性抑制剂为对象,研究基于靶蛋白结构的配体合理设计和组合设计、高通量筛选、生物学评价、潜药及软药设计;

(3) 以尿酸酶、芳香硫酸酯酶和碱性磷酸酶为对象,研究酶突变体合理设计和定向进化、酶突变体高通量筛选、酶突变体应用评价、蛋白质化学修饰制剂;

(4) 药物靶蛋白亲和标记和位点选择性固定化技术、抗体的酶标记技术;
